This excellent scientific article by the top mitochondrial expert shows how so many of our systems can be so severely affected.
From microlglia, to mitochondria. It is all about CELL SIGNALING.

How can we get this to NORD? This knowledge I believe will be essential in the resolution of Pois, autism, cfs and many others.

This is an immensely important set of scientific knowledge.

Naviaux's got some big accomplishments behind him and some big toys to play with. His lab developed two advanced technologies; biocavity laser spectroscopy and mtDNA mutation detection by mass spectrometry, and recently created new bioinformatic methods that allow them to better analyze genetic data. Put all that together and Naviaux says he has the ability to "dissect the metabolic and molecular features of virtually any disease".

Interesting new theory, Mrraba, but it will take some time to really grasp all it implies.  I will look more into it. It will take some time for me to study it, since it covers a large array of metabolical events.  But, just looking at the B section of figure 2, showing 9 metabolic processes with their ill or healthy state, is very striking - many, if not all, of those nine process have been discussed on the forum  ( gut microbia, mehtylation, kynurenine production via tryptophan pathways hacking, etc...).  Needless to say that it is an ambitious synthesis of what is causing "chronic metabolical illnesses", or the like those resisting modern medecine so far  ( including  CFS/ME, and POIS !)


The old pathways mentioned, those controlling the so-called CDR, are not without reminding me of the "old vagus" response mentioned in the polyvagal theory, and could be one and the same, viewed from different angles.

Interesting enough, the article states that lysine supplements opposes CDR, and I have some lysine coming in my current supplements order ( i wanted to test its anxiolytic properties).  However, I would never, never take Suramin, the drug used by Dr Naviaux ( mainly on rats, I think, hopefully), it is a very dangerous drug, with very severe side effects  ( including sudden deaths, leading to stop using it in HIV patients ).  I prefer, to say the least, Lysine, a naturally occurring amino acid, or safe, low doses of P5P ( vitamin B6) :)

After 23 years of sudden POIS and CFS onset I have heard of many things that help some, diverse things like low sugar diet, meditation, immunocal  ( powerful effect almost instantly ),  magnesium,etc. Yet a perspective that would unify these approaches was elusive. It is just very complex. All our systems are involved.  Anyone with POIS or CFS can attest that the impact feels  systemic.

This perspective fits our symptoms and the state of current Knowledge  of autonomicd/metabolic disorders so well.

I agree with you that the vagus nerve is involved. As a meditation/mantra practitioner I have experienced the deep effets these have in CNS balance.  Perhaps the vagus nerve is just one of many key functional systems affected. In my opinion the vagal nerve is 100% in this. But my feeling is that the cause of its involvement is much deeper and widespread. Cell signaling seems a good candidate for involvement.  Definitely a meaty and very rich Article. The amazing thing is that theory as is, resulted in full remission and reversal of autism in mice. Absolutely incredible and hope raising. Yes do not even think about tryng the drug used for that. I expect there will be better ones.

What it says on gluten, multiple sensitivities, and AMPK is striking to me. I have a relative who recovered from CFS with AMPK amplifier Merformin. But no one knew why. Effect on AMPK possibly.  Also low carb  diets  boost  AMPK.   But  Metfformin disabled me temporarily. It was awful.  So now  I know it was because it increases lactic acid.
