METHYLENE BLUE!!!!
« on: November 20, 2014, 02:04:17 PM »
methylene blue acts directly on the nervous system, where you notice the problems. The nervous system governs the intestine, inflammation, hormones, metabolism, everything. You'll have to play around with the dose and see what works for you. I took 3-4mg yesterday and to my pleasant surprise, NO POIS. I'll keep you guys updated if this continues to work. There is a big thread on longecity forum about it if you want to check it out, but to my understanding, those guys are waaay underdosing it.

so the thought of the experiment was a scary one, like it would be for any POIS sufferer, orgasm few times a day for a few days to see what happens. After that experiment I can conclude, I have zero POIS while taking this stuff. I jizz, and then feel the exact same right after.. there might be a slight cognitive decline, but its maybe 5% at most.

Wow. Where can I buy this?
Complete loss of focus, extreme fatigue, sweating, nasal irritation, joint pain, headaches and depression. Takes a full week to feel normal again. Hiding from life. Praying for a cure.

Re: METHYLENE BLUE!!!! - ***** DRUG AND FOOD INTERACTION WARNING !!!
« Reply #3 on: November 24, 2014, 01:52:47 AM »
Hi, Pois_Dick, and everyone,

As a pharmacist, I would like to make an IMPORTANT WARNING regarding possible major, and potentially fatal, interactions of methylene blue with many prescription drugs boositng serotonine level, with tryptophane supplements, and also with tyramine-containing food.

Most likely, the benefits of ingested methylene blue you have seen may come form the fact methylene blue is a potent monoamine oxydase inhibitor (MAOI), so it increases serotonin level.  I  would like to point out that ANYONE WHO WANTS TO INGEST METHYLENE BLUE MUST BE VERY CAREFUL !!!!  IT CAN BE VERY TOXIC, AnD EVEN DEADLY IF INTERACTIONS OCCUR WITH DRUGS ALSO BOOSTING SEROTONINE LEVEL AS WELL AS WITH FOOD CONTAINING TYRAMINE !! See https://en.wikipedia.org/wiki/Monoamine_oxidase_inhibitor#Dangers

With tyramyne containing food, like aged cheese, wine, Bovril/Marmite\Vegemite meat extract, a severe hypertensive crisis can occur, possibly fatal. See a list of fod containing tyramine here:  https://en.wikipedia.org/wiki/Tyramine#Occurrence .  If you take MB, you must avoid them, unless you will trigger a massive norepinephrine release in yout body, leading to hypertension, anxiety, etc...

BUT there is an even worst risk - IMAO like Methylene blue interacts with Tryptophane supplements and with many drugs POISers on this board are taking. From Wiki?dia:" MAOIs should not be combined with other psychoactive substances (antidepressants, painkillers, stimulants, both legal and illegal etc.) except under expert care. Certain combinations can cause lethal reactions, common examples including SSRIs, tricyclics, MDMA, meperidine,[30] tramadol, and dextromethorphan.[citation needed] Agents with actions on epinephrine, norepinephrine, or dopamine must be administered at much lower doses due to potentiation and prolonged effect."

Form wikipedia again: "The most significant risk associated with the use of MAOIs is the potential for interactions with over-the-counter and prescription medicines, illicit drugs or medications, and some supplements (e.g., St. John's Wort, tryptophan). It is vital that a doctor supervise such combinations to avoid adverse reactions. For this reason, many users carry an MAOI-card, which lets emergency medical personnel know what drugs to avoid. (E.g., adrenaline dosage should be reduced by 75%, and duration is extended.)[27] The risk of the interaction of MAOI medications with other drugs or certain foods is particularly dangerous because those on the medication who would have to restrict their diets often are depressed patients who "don't care if they live or die."[28]

Tryptophan supplements should not be consumed with MAOIs as the potentially fatal serotonin syndrome may result."

So, DO NOT USE METHYLENE BLUE WITHOUT MEDICALAND PROFESSIONNAL SUPERVISION, AND WITHOUT CHECKING FOR POTENTIAL INTERACTIONS AND WITHOUT KNOWING ABOUT FOODS TO AVOID !!

And, if you still want to use it, be very careful and go with minimal dosage (as with any treatment, go for the minimum efficient dosage), though i recommand not to use it at all.

I do not want to scare anyone, but want you to stay safe !

my guess isn't because of the MAOI effect, but because it delays cellular senescence and enhances key mitochondrial biochemical pathways, especially in the brain. It is dangerous to take in combination with SSRI's, but I have not read anywhere about the toxicity you describe. In fact, it was shown to be safe at several grams/day.

Hi, Pois

In addition to the links already given in my previous posts, here are 2 other links, from the FDA, and from the Mayo Clinic:

The Drugs - Methylene Blue (MB) interactions are well documented:

https://www.fda.gov/Drugs/DrugSafety/ucm276119.htm

Tryptamine supplements are also interacting badly with MB

Based on the proven fact that MB has MAOI properties, the well known interactions with Tyramine-containing foods apply as well to MB, but tyramine content of food vary greatly, and personal tolerance from one person to the other differ also, so there is no clear guidelines as to how much of this and that food can be taken with this much of that much of an MAOI, including MB, without risk of an hypertension crisis.  For more details:

http://www.mayoclinic.org/diseases-conditions/depression/expert-answers/maois/FAQ-20058035

Moreover, the data for oral intake do not really exist, because MB is used in IV injections, in current medecal practice, so there is no data on the bioavailability of MB, that is, the % of the orally taken dose that will make it to the blood stream.

Take care!

Methylene Blue Success?
« Reply #6 on: January 05, 2024, 05:14:50 AM »
I have gotten extreme benefits from methylene blue in regards to every single symptom I have (physical, mental, cognitive), and it works very quickly. I have tried countless supplements, all of which have had zero impact on my symptoms except for methylene blue. Any ideas on why this might be? I should mention that when I abstain for at least a week and do an extended water fast I get the same exact effect that I do from methylene blue. I feel like there is a connection there but I don't know what it would be. Does anybody have a similar experience with MB?

It may be that some toxic elements irreversibly bind with hemoglobin in your blood forming methemoglobin, which makes it unable to transport adequate oxygen. This condition is called methemoglobinemia and methylene blue may be used as a reversal measure.
https://en.wikipedia.org/wiki/Methemoglobinemia
Methylene blue also proved beneficial in sepsis treatment, which also shares some features of POIS.
https://en.wikipedia.org/wiki/Septic_shock

As I see methylene blue is mostly available in liquid form as a fish tank additive that may help the fishes to overcome infections. Alternatively it is sold in liquid or solid form in bulk for laboratory use.
Which form do you use and in what dilution and quantity?
The cause is probably the senescence of sexual organs and resultant inducible SASP, which also acts as a kind of non-diabetic metabolic syndrome.

I use liquid USP grade methylene blue (https://www.amazon.com/Methylene-Blue-Pharmaceutical-Grade-Twin-Pack/dp/B0BNK6KQ19/ref=asc_df_B0BNK6KQ19/?hvadid=653103648180&hvpos=&hvnetw=g&hvrand=7349130383169078143&hvpone=&hvptwo=&hvqmt=&hvdev=c&hvdvcmdl=&hvlocint=&hvlocphy=1026899&hvtargid=pla-2111299266127&psc=1&mcid=c62a5886ce18302db72e79ab304f1635)

I use 10 drops which is equal to about 5 mg. I just drop them into a glass of water. My first guess was that it was addressing mitochondrial deficiency, although I have tried CoQ10 and NAD+ to no avail. The conditions you mentioned are interesting, although I don't think they really align with what's happening. The similarities in from my experience with MB and water fasting lead me to believe that it does have to do with increased oxygen flow, but if it's not mitochondrial deficiency or methemoglobinemia I don't know what it would be or why it is ejaculation, food sensitivities, exercise etc. would be making it worse.

Hi Phantom,

Chances are that methylene blue helps you because it is a potent monoamine oxydase inhibitor (MAOI), which is a class of antidepressants that are no more used today because of the dangerous side effects they could have, and their high potential for interaction with over-the-counter products and certain food.

So, you should read the post I made almost 10 years ago, above on this thread (https://poiscenter.com/forums/index.php?topic=1551.msg14553#msg14553  ).  You should also read the sections at https://en.wikipedia.org/wiki/Monoamine_oxidase_inhibitor#Side_effects .

In a nutshell, MAOI in addition to food containing tyramine,  like  cheese, Chianti wine, and pickled fish, can lead to a severe hypertension crisis.

Also, The most significant risk associated with the use of MAOIs is the potential for drug interactions with over-the-counter, prescription, or illegally obtained medications, and some dietary supplements (e.g., St. John's wort, tryptophan).  These interactions can lead to what is called a serotonin syndrome   ( Serotonin syndrome (SS) is a group of symptoms that may occur with the use of certain serotonergic medications or drugs. The symptoms can range from mild to severe, and are potentially fatal. Symptoms in mild cases include high blood pressure and a fast heart rate; usually without a fever. Symptoms in moderate cases include high body temperature, agitation, increased reflexes, tremor, sweating, dilated pupils, and diarrhea. In severe cases, body temperature can increase to greater than 41.1 °C (106.0 °F).Complications may include seizures and extensive muscle breakdown.)

So, if you really want to go on with methylene blue, stay away that anything that could interact with it, like antidepressants drugs, and be sure to consult a health professional about it.  The dose you mention, about 5mg a day, is low, but be sure to stay on the safe side !

Still the up and down roller coaster of Pois phenomenon.
Some symptoms of tyramine  excess look like first phase of Pois. Then MB might help with the 2nd phase by supplementation of depleted cathecolamines.

Quantum, THANK YOU! Your very important  and expert summation [note: Quantum is a professional pharmacologist] certainly stopped me in my tracks from rushing out and trying Methylene Blue - - without most careful consideration of the risks!

Your risk-downside comments certainly caught my eye:

“…dangerous side effects they could have…”

“…can lead to a severe hypertension crisis.”

“The symptoms…are potentially fatal.”

Thank you again!

Started with Methylene Blue (24-1-24). Saw the thread on poiscenter and actually know a guy who is into hacking health (is healthy himself) and is using MB with red light therapy because of its properties. 1 gram MB powder (CZTL) on 100 mL water. I'm taking 10 drops (5mg) in the morning and 10 drops before sleep. No issues so far.

It seems to have a blocking effect on the Vagus nerve: https://link.springer.com/article/10.1007/BF00781015
Book about MB:
The Ultimate Guide to Methylene Blue: Remarkable Hope for Depression, COVID, AIDS & other Viruses, Alzheimer’s, Autism, Cancer, Heart Disease, Cognitive ... Mitochondrial Dysfunction) https://annas-archive.org/md5/258e416ff2e1482108a75667152ecea8

Edit:
Wow. Where can I buy this?
CZTL

Tip: if you buy a 100 mL bottle then you will not have to measure anything, just fill it all the way up and add the content of the 1 gram vial (flush the vial for the residu that sticks to the wall). Perhaps you want to add the powder outside of the bottle to the water to mix it properly before throwing it back in.

Important note: MB is photosensitive, thus needs to be stored in dark tinted glass.

March 2, 2024. Still using the same dose daily, which is relatively low. Cannot say it affects me in a positive or negative way. A higher dose is used in research. I might increase the dose and/or frequency, I have not decided yet. It accumulates within the brain without inducing symptoms (blue brain).

I gave some Methylene Blue of the same brand to my younger brother. He had sent me a message that the stuff works immediately even at low dose. He didn't specify which symptoms improved nor the dose. I will get back to this.
