Cured POIS and possible CYP24A1 Connection

I have recently cured my POIS with niacinamide (500 mg daily), liposomal vitamin C 700 mg daily (with phospholipid complex from sunflower oil and lecithin), and magnesium malate (100 mg daily) supplementation. Here is some background on myself. When I did this all of my issues regarding not only POIS but MCS (multiple chemical sensitivity) and EMHS (electromagnetic hypersensitivity). I also used to have a sensitivity with metals that was far worse when I worked at a job where I was handling large amounts of change every day resulting in burning feeling in my face and changes in state of mind. I also have food sensitivities like lactose intolerance, gluten intolerance, and acidic foods like tomato sauce causing psychiatric issues shortly after consuming that can last for up to a day after including mania, depression, and higher levels of aggression. I am not sure if I have all of these problems anymore but based on my personal diet and what i have been eating I am more mentally functional now after eating bigger meals and no longer have exaggerated mental disturbances upon consuming anything so far. I have not tried any gluten, tomato sauce, or something that would be a real test pizza yet but I plan to eventually and take note of the results.

Something was obviously sped up and very efficiently. In looking up other things related to liver enzymes I found a connection to the CYP24A1 enzyme specifically. I believe many with POIS have a mutation in this enzyme.

The Cytochrome P450 is involved with the metabolism of many drugs and other toxins/carcinogens. When this what I believe to be a block in this system was resolved everything improved.

What led me to this conclusion is that in a Discord server about POIS many member have had elevated calcium in urine (hypercalciuria) including myself all of our lives. I haven't been to a doctor and haven't seen one in a long time nor do I plan to as I independently take care of myself currently but this has always been the trend from all past visits in my life. Another common association was elevated liver enzymes, what these were I can't remember as I was never told a long while back unfortunately which I can't find any direct study showing association between this and CYP24A1 but was a common thread anyways. This (hypercalciuria) is associated with CYP24A1 dysfunction from a mutations. I am also on the spectrum (ASD) and again this may also be linked to dysfunction of this enzyme to further back up what I am claiming. There was no data presented here as it was all word of mouth (or text by the matter) so it must be taken as is.

On a personal note that may also indicate something going wrong, if I do not supplement with vitamin D my liver seriously struggles one time resulting in the yellowing my skin and eyes to a degree when I once decided to see what would happen if I stopped taking it along with severe fatigue and migraines. This was done many years ago and I do not know if taking it out of my every other day supplementation of 5000 IU every other day would produce the same results now. At the time this happened my MCS and EMHS was at an all time peak too.

"Haplotype analysis revealed several potential regions with p-values ranging from 1.8*10−4 to 3.6*10−6 (S1 Table); haplotypes in genes NAALADL2, SGSM2/MNT, and CYP24A1/BCAS1 showed associations with ASD. For gene-set analysis, we included 314 genes with p<0.01 from the GWA analysis and results are listed in Table 2. We found 14 significant pathways for ASD such as the olfactory signaling pathway, G-protein coupled receptor (GPCR) downstream signaling, cytoplasm, and cell proliferation."

I haven't tried this yet but I plan to do so in the near future.

Doing more research I found that estrogen is partially metabolized in the liver by CYP24A1 in extrahepatic tissues. There is also the high possibility that i have other mutations in this enzyme family too - https://pubmed.ncbi.nlm.nih.gov/16112414/

CYP24A1 dysfunction pointing towards a reduction of function and toxic and/or estrogen build up in the body due to a possible hormone recycling problem may be the key in curing people with POIS. It is also worth looking into other mutations in the Cytochrome P450 family or other closely related systems.

Wow great work on discovering your cure and potential underlying root cause, we are getting closer with each day to solving this evil disorder!

Is this still working for you?  Are we talking 100% symptom relief or partial relief?

Mindlesstree, this is quite fascinating information.  I also take daily low dose vitamin D and C, which moderately helps with lingering symptoms. Please update us on your road to recovery!
