The approach of POIS as simply an allergy to one own sperm is no more in line with the latest work of Dr Waldinger, the one who first brought this skin prick testing and hyposensitisation technique. Now, Dr Waldinger speaks about a complex immune reaction - read all the details in the thread at

In short, some men with positive skin testing do not have POIS, some with negative testing do have POIS, so the test is inconclusive. Dr Waldinger do not use it anymore, and have abandoned desensitization injections as well. He even apologize for the confusion about the "allergy" term, recognizing that he may be responsible for it. An auto-immune reaction, which is more complex than a simple allergic reaction, cannot be diagnose with prick tests, and cannot be treated with desens. Unfortunately, some internet articles about POIS still talk about it as an "allergy" and still cites desens injections as a treatment.
For now, there is no objective test that can diagnose POIS, only the clinical presentation, with the 5 criteria established by Dr Waldinger, can be used as a guideline for POIS diagnostic. For the exact 5 criteria, set by Dr Waldinger, see

In his current study, Dr Waldinger is looking for the mechanism of the auto-immune reaction in POIS, so he may come up with a diagnostic test for it, if he succeed in finding the antigen and auto-immune mechanism he believes is at the core of POIS.
