Title: Neurosteroids are involwed in POIS mechanism
Post by: Hopeoneday on July 11, 2019, 09:26:38 AM
While i resarching why benzodiazapines in small doses helped
me to surviwe In POIS atac, i dicovered that Neurosteroids are
wery potent positive GABAa receptors modulators.
(mainly by Allopregnanolone , androstanediol...)

So, syntesis of neurosteroids
in brain for exemple depend from pregnelone, progesterone,
testosterone by enzyme pathways...

List of neurosteroids.

Allopregnanolone

https://en.wikipedia.org/wiki/Allopregnanolone

Allopregnanolone possesses a wide variety of effects, including,
in no particular order, antidepressant, anxiolytic, stress-reducing,
rewarding,[9] prosocial,[10] antiaggressive,[11] prosexual,
[10] sedative, pro-sleep,[12] cognitive, memory-impairment
....

Increased levels of allopregnanolone can produce paradoxical effects,
including negative mood, anxiety, irritability, and aggression.[17][18][19]

I think you are close to the truth and  GABA receptors is a key of the mystery.

I think you are close to the truth and Cholesterol the pre-cursor to Neurosteroids may be playing a key role in POIS :)

Excellent post HOD, neurosteroids are definitely a strong candidate. Maybe that's why dexamethasone works for me.

After 5 ejaculations, my bloodtest showed :
-normal total testosterone
- normal but low limit free testosterone
- DHEAs too high

An old bloodtest (15 years?) showed low total cholesterol out of Pois.

DHEAs too high would give me "male" symptoms : muscles, hairy strong voice... that I doen't have.

My total cholesterol has always been low. You can also find members on this forum who have high levels which makes a potential link between POIS and cholesterol questionable. Are there any tests available that can verify this theory? What neurosteroid could be tested?

I also like to add, are neurosteroids endogenous in the brain or do they affect the brain when produced ( outside of the nervous system )?

Muon, FWIW, I’ve been on cholesterol meds (rosuvastatin calcium aka Crestor) since open heart surgery.

One contradiction with this theory is that both inhibitory and excitatory neurosteroids have benefits; the former is  anti-depressant, anti-anxiety, pro-social, pro-sleep, etc. The latter is pro-memory and pro-cognitive. This can not fit into POIS, since the majority of POISers experience both cognitive and psychological symptoms, even those who have agitation symptoms also report cognitive issues. I don't know how you can amend this contradiction since you can't have only the negative effects without the positive effects.
